Spider-Man: Across the Spider-Verse features a variety of dimensions, and the Marvel Cinematic Universe (MCU) could be some of them.
Chris Miller revealed more details about the ambitious project on the Happy Sad Confused podcast. During the interview, Miller teased a potential connection to the Marvel Cinematic Universe. When asked about the possibility of connecting to the live-action Spider-Man movies, Miller didn't rule out the chance.

Mainly, Miller mentioned that the Multiverse is a medium through which this could occur.
The Multiverse is big and wide. And all things grow... Why would you think a Multiverse, in which many things are possible, that [those things are] not related?
click to enlargeVaried characters were captured in the Oscar-Winning film, as Miles was joined by the black-and-white Spider-Man Noir and a more cartoonish Spider-Ham. Peni Parker accompanied Spider-Gwen too, so it's certainly possible this ensemble will expand further.
Whether these ties will exist as actual characters or simple easter eggs to Tom Holland's world, we shall wait and see. However, considering the multiverse was pulled off in No Way Home, Spider-verse could expand upon that by re-introducing the three Spider-Men.
Spider-Man: Across the Spider-Verse's ambitious animation is matched by its story, with Part One slated for this year and Part Two in 2023.
These sequels feature the return of Shameik Moore as the leading hero, along with Hailee Steinfeld's Gwen Stacy and Jake Johnson as Peter B. Parker. Oscar Isaac and Issa Rae will be joining the cast as Spider-Man 2099 (Miguel O'Hara) and 1977 Spider-Woman (Jessica Drew) respectively.
